Increased plasma leptin levels were associated with increased insulin resistance, inflammatory markers and haemostatic risk factors for cardiovascular disease in 3640 non-diabetic men, 60 to 79 years of age, drawn from general practices of 24 British towns. The association remained significant after adjustment for waist circumference and lifestyle factors such as physical activity and cigarette smoking, hinting at potential pathways by which leptin may influence cardiovascular risk. Leptin may contribute to CVD as a result of its effect on IR, as well as through its pro-inflammatory and prothrombotic actions, and may therefore represent one of the “missing” links between obesity and CVD.
